Bacteria labeling with GFP? help?

Wow there’s a lot to unpack here.

First and foremost, you need to talk to your PI about this because it’s a full ass masters project or a chapter in a PhD. Ideally, this would take you maybe 6 months to 1 year, but realistically you’re going to stumble through this project and mess up often (especially if this is your first time cloning, which seems to be the case) which will delay the project further. Each step of this would be incredibly complicated and you would need multiple meetings to go over protocols, troubleshooting, and results that you may find.

To answer some of your questions (which your PI would be way more qualified to answer):

-To tag a specific organ, you’ll most likely need a plasmid with a tissue-specific promoter such as lung, neuron, muscle, etc. -You’re probably lacking some key information since you wouldn’t be just tagging an organ with GFP for no reason. Ask if there is a specific component that you’re targeting within the organ (most likely a protein). -Your plasmid requires a few components such as a selectable marker gene (antibiotic resistance), promoter, origin of replication, and a multiple cloning site.

You should definitely read up on different cloning strategies and discuss them with your boss. It’s pretty important to understand the basics of what you’re doing so you can follow a clear path with logical steps. As a side note, I don’t mean to come off as condescending or rude, but this is a surprising amount of work to be asking about online.
