Help! My 4Runner is dead in the water!

The 22re's have a weird way of powering the fuel pump. It goes through the AFM to get power to the pump. If the engine turns over and the AFM doesn't move from the vacuum caused by the engine then the pump never gets its power and therefore won't send fuel to the rail. I had a similar issue very recently. My AFM was corroded and the flapper\door wouldn't move at all. Took it apart and cleaned it up with electronics cleaner and lubed the pivot point and she fired right up after reassembly.

If spraying starting fluid in the intake manifold will make the engine start but not continue to run then you're not getting fuel. Check the entire fuel pump circuit, to include the AFM.
